The Audiophile does not have a built in wavetable, which is the actual audio sounds that the MIDI data turns in to. The Audiophile only passes the data in and out of it to recieve it from one device and send it to another.

I don't use Reason, so I can't speak for that, but VST and DX instruments generally use samples of some sort, such as wave files, mp3 files , sound fonts or things of that nature to turn the MIDI data into sound...therefore, your controller keyboard should work fine in those cases.

I had the Audiophile and recently switched to the ESI Juli@ and it has the same issue regarding no wavetable. My Radium49 however, still works fine by just triggering audio samples from within the computer itself.

the audiophile will work with VST instruments as long as the samples are bundled into the program. reason will work. but there may be settings to make in places you might not suspect. i figured it out just by systematically adjusting everything but i no longer remember what the specific problems i had were. keep playing with it, post any settings and your progress here and you should be able to get it working.
