The VSTi's can reside on your main drive without any problem. They run natively inside Cubasis and won't cause any less stress on the CPU if they are moved to a different folder on another drive.
That said however, if you have a sampler type VSTi that uses large wav files or other audio type files, those can be moved to another drive for better results and speed. But the catch is that they need to be directed there when installing the VSTi itself. I use several types of VSTI and DXi that require large sample files and that is how I do it. Also that is where I store all my project and audio files from each project. That is the biggest thing you can do to relieve some of the burden on the OS drive.
I do know that there are some instruments that will allow you to re-direct the sample and wave files they use to another folder, but it is dependent on each different VSTi as to how they require you to do it.
If you have a list of them I may be able to give you an idea on how to do it if I am familiar with the particular VSTi.
