Confusion with buses

This seems to me like a complicated question but the answer is probably simple. I'm having some trouble on how to set up my buses and sources for recording. My setup is a small yamaha mixer going into a m-audio 2496 and then into adobe audition. my question is whether, when recording in mono, the setting of the bus on the mixer matters, and if so, where it should be. Also, does it matter if i record in mono or stereo if im only recording with one mic? What i've been doing has worked so far, but im wondering whether some fine tuning will improve my results.
