Are they samples of single note hits of instruments? Or are them loops of instruments? Do you have a sampler software to use in Nuendo?

These answers will help get you going.

I believe Nuendo uses a standard piano-roll to play midi files. You'll need to work out how to put those sounds into a midi-map. Is your bank of sounds like a soft-synth?
If your sound card has a midi input, you should be able to control with a midi keyboard.
Other than that, there are some free VSTi samplers out there.

Steinberg's (Cubase/Nuendo) manuals are usually quite well written, so you should be able to go step by step to get there.

Good luck.

Oh, and watch buying software and stuff on eBay. Some manufacturers don't allow for transfers of user without a fee.

What instrument package did you buy? It may have come with its own player depending on what exactly it is.
